Induction of Premature Chromosome Condensation (PCC) by Calyculin A for biodosimetry

چکیده
B iological dosimetry of ionizing radiation is generally performed by scoring dicentrics and/or translocations in chromosome from peripheral blood lymphocytes (pbl) at the first mitosis following in vitro growth stimulation. It is well known that after irradiation, in this procedure mitotic index is low in some individuals, so the scored mitoses might not be representative of the exposed cell population. For these reasons premature chromosome condensation (PCC) was introduced by Johnson and Rao (1970) following fusion of an interphase with a mitotic cell (Johnson and Rao 1070) 1970). With the use of this method makes it is possible to distinguish individual chromosomes at different stages of the cell cycle. Thus, it has been used to study the formation of chromosomal aberrations shortly after irradiation and the kinetics of chromosome break rejoining (Pantelias and Maillie 1985). This technique is considered as a potent biodosimetric tool (Blakely et al. 1995) since it is probably one of the most sensitive methods for analyzing initial chromosomal damage after irradiation (Sipi et al. 2000). At present, PCC can be induced by two methods, cell fusion and using chemicals. In cell fusion method, CHO mitotic cells with high promoting factor (MPF) are fused to interphase cells by polyethylene glycol (PEG) or Sendi virus. After staining, chromosome elements appear as single chromatids and can be discriminated easily from the CHO mitotic chromosomes in human interphase G0/G1 PCC spread Induction of Premature Chromosome Condensation (PCC) by Calyculin A for biodosimetry

Induction of premature chromosome condensation (PCC) by calyculin A for biodosimetry
Background: Premature chromosome condensation (PCC) is a method for interphase chromosome analysis in biodosimetry. This study was performed to verify the usefulness of PCC induced by calyculin A in human peripheral blood lymphocytes (pbl) for biological dosimetry and possible construction of dose-response curve. Materials and Methods: Peripheral blood was obtained from a healthy donor and expo...
متن کاملBiological Dose Assessment by Chemical Induction of Premature Chromosome Condensation
Dose assessment of radiation victims is a key element in medical management of radiation accidents. At high radiation doses (> 6 Gy), dicentric assay lose its efficiency and premature chromosome condensation (PCC) assay was proposed to overcome the restrictions of dicentric assay. It was shown that PCC ring is more suitable and simpler than dicentric for biodosimetry in high radiation doses. In...

Premature Chromosome Condensation (PCC) appears to have a possible utility for biological dosimetry purposes. The PCC technique may be adapted for cases of suspicion of overexposure where sampling is performed at least one day after an accident. For this purpose, human blood samples were exposed in vitro to 60Co up to 10 Gy and the PCC technique was performed immediately after irradiation. Anal...

The combination of automatic image acquisition and automatic image analysis of premature chromosome condensation (PCC) spreads was tested as a rapid biodosimeter protocol. Human peripheral lymphocytes were irradiated with (60)Co gamma rays in a single dose of between 1 and 20 Gy, stimulated with phytohaemaglutinin and incubated for 48 h, division blocked with Colcemid, and PCC-induced by Calycu...
متن کاملA comparison of G2 phase radiation-induced chromatid break kinetics using calyculin-PCC with those obtained using colcemid block.
To study the possible influence of cell-cycle delay on cells reaching mitosis during conventional radiation-induced chromatid break experiments using colcemid as a blocking agent, we have compared the chromatid break kinetics following a single dose of gamma rays (0.75 Gy) in metaphase CHO cells using calyculin-induced premature chromosome condensation (PCC), with those using colcemid block. Ca...
